Gatifloxacin mesylate works by inhibiting the activity of bacterial DNA gyrase and topoisomerase IV. These enzymes are essential for bacterial DNA replication and transcription. By inhibiting these enzymes, gatifloxacin mesylate disrupts bacterial DNA synthesis, leading to cell death.

This mechanism of action makes gatifloxacin mesylate effective against a wide range of bacteria, including those that are resistant to other antibiotics. It has activity against common pathogens such as Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, Staphylococcus aureus, and Streptococcus pneumoniae.

After oral administration, gatifloxacin mesylate is rapidly absorbed from the gastrointestinal tract. It has good bioavailability, with approximately 96% of the drug being absorbed. Peak plasma concentrations are reached within 1 to 2 hours.
